Lateral Flow Test (LFT) Detects Cell‐Free MicroRNAs Predictive of Preterm Birth Directly from Human Plasma

Despite extensive research toward the development of point-of-care nucleic acid tests (POC NATs) for detection microRNAs (miRs) from liquid biopsies, major hurdles remain including strict requirement off-chip sample preprocessing. Herein, a lateral flow test (NALFT) is reported on that enables direct endogenous miRs as little 3 μL plasma without any enzyme-catalyzed target amplification or complex miR extraction steps. This achieved through integration denaturing hydrogel composite material onto LFT, allowing near-instantaneous on-chip release their carriers (extracellular vesicles transport proteins) prior to detection. next-generation LFT sensitive enough detect concentrations miR-150-5p, predictive biomarker preterm birth (PTB) found deregulated in maternal blood early 12th week pregnancy. key step represented first bedside risk-stratification during pregnancy by predicting true outcome at very stage. More generally, universal and versatile nature this novel preprocessing platform can further improve robustness existing NALFTs facilitate application POC.
